Utilization of agro-industrial by-products as substrates for dextransucrase production by Leuconostoc mesenteroides T3: Process optimization using response surface methodology

Dextransucrase (DS) is a glucosyltransferase (E. C. 2.4.1.5) that catalyzes the transfer of glucosyl residues from sucrose to dextran polymer and liberates fructose. This enzyme isassociated with wide application range oligosaccharides. DS production by Leuconostoc mesenteroidesT3 was optimized using Central Composite Design under Response Surface Methodology. Three variables were chosen for optimization: distillery stillage, manganese concentration. The results showed concentrations had positive linear effect on while all variable interactions (stillage-manganese, stillage-sucrose, sucrose-manganese) significant influences production. maximal yield 3.391?0.131 U cm-3, obtained in medium 64.33 % stillage concentration, 5.30% concentration 0.022 Our study revealed potential combined sugar beet molasses, supplemented be employed as valuable growth lactic acid bacteria DS. Also, taking into consideration origin substrates, utilization industrial by-products this way has great environmental relevance accordance circular economy.
